چکیده

Phage typing is an important tool to discriminate the strains of Vibrio cholerae from epidemiological point of view. National Institute of Cholera & Enteric Diseases is engaged in research on cholera phages since decades in India. Bacteriophages have long been used for classification and discrimination of bacterial strains. Cholera, an endemic disease in Indian subcontinent with mild to lethal outcome has been studied in detail using a set of bacteriophages sensitive to a particular subtype to classify V. cholerae. So, bacteriophage typing of Vibrio cholera was an extremely important tool before few decades and still today it is very important. Other than typing, bacteriophages can be used as therapeutic and the field of phage therapy attains high importance in recent years. Phage therapy experiment was found effective in animal model where we found that the combination of five phages was highly effective and did nonetheless reduce the severity of cholera diarrhea. This novel approach will be further pursued by utilizing cocktail phage in preclinical and clinical trials. Our future strategy is to carry forward our mission by utilizing the cocktail phage in human volunteer study.

Structure of vibriophage D10 tail sheath revealed by electron microscopy and computational image processing.

Vibriophage D10, a member of the Vibrio cholerae O-1El-Tor phage typing scheme, is used to detect the spread of the cholera epidemic and belongs to the Myoviridae family. The outer sheath of the tail of vibriophage is highly contractile in nature. We have used electron microscopy and computational image-processing techniques to determine the structure of this contractile tail sheath. The three-...

Vibrio cholerae typing phage N4: genome sequence and its relatedness to T7 viral supergroup.

OBJECTIVES In countries where cholera is endemic, Vibrio cholerae O1 bacteriophages have been detected in sewage water. These have been used to serve not only as strain markers, but also for the typing of V. cholerae strains. Vibriophage N4 (ATCC 51352-B1) occupies a unique position in the new phage-typing scheme and can infect a larger number of V. cholerae O1 biotype El Tor strains. Here we c...

Modeling the role of bacteriophage in the control of cholera outbreaks.

Cholera is a waterborne diarrheal disease that continues to plague the developing world. Individuals become infected by consuming water from reservoirs contaminated by virulent strains of the bacterium Vibrio cholerae. Epidemiological and environmental observations of a cholera outbreak in Dhaka, Bangladesh, suggest that lytic bacteriophage specific for V. cholerae may limit the severity of cho...

Non-toxigenic environmental Vibrio cholerae O1 strain from Haiti provides evidence of pre-pandemic cholera in Hispaniola

Vibrio cholerae is ubiquitous in aquatic environments, with environmental toxigenic V. cholerae O1 strains serving as a source for recurrent cholera epidemics and pandemic disease. However, a number of questions remain about long-term survival and evolution of V. cholerae strains within these aquatic environmental reservoirs. Through monitoring of the Haitian aquatic environment following the 2...

Mechanisms of the evolutionary arms race between Vibrio cholerae and Vibriophage clinical isolates.

This review highlights recent findings on the evolutionary arms race between the causative agent of cholera Vibrio cholerae and virulent bacteriophages (phages) ICP1, ICP2, and ICP3 isolated from cholera patient stool samples. We discuss mechanisms of phage resistance such as a unique phage-inhibitory chromosomal island and mutations that affect phage receptor expression. We also discuss the mo...
